We aimed to characterise the kinetics of maternally and naturally acquired antibodies against historical A(H1N1)pdm09 strains and to assess the extent to which the response to heterologous strains following infection or vaccination affects observed A(H1N1)pdm09 strain-specific antibody titres in a Chinese paediatric population.<\/p>\n\n\n\n<p>\u80cc\u666f 2009 \u5e74\u65b0\u578b\u7532\u578b H1N1 \u6d41\u611f\u75c5\u6bd2\u51fa\u73b0\u4ee5\u6765\uff0c\u5176\u6297\u539f\u53d8\u5f02\u9891\u7e41\uff0c\u6301\u7eed\u5f15\u8d77\u513f\u7ae5\u7684\u4e25\u91cd\u611f<br \/>\u67d3\u3002\u672c\u7814\u7a76\u65e8\u5728\u63cf\u8ff0\u7532\u578b H1N1 \u6d41\u611f\u75c5\u6bd2\u7684\u6bcd\u4f20\u548c\u81ea\u7136\u611f\u67d3\u8bf1\u5bfc\u6297\u4f53\u7684\u52a8\u529b\u5b66\u7279\u5f81\uff0c\u5e76\u8bc4\u4f30<br \/>\u81ea\u7136\u611f\u67d3\u6216\u75ab\u82d7\u63a5\u79cd\u5bf9\u7532\u578b H1N1 \u6d41\u611f\u75c5\u6bd2\u6bd2\u682a\u7279\u5f02\u6027\u6297\u4f53\u6ef4\u5ea6\u7684\u5f71\u54cd\u3002<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Methods<\/h3>\n\n\n\n<p>In this retrospective study, we used residual serum samples from 528 mother-neonate pairs from a non-interventional, longitudinal cohort study in southern China conducted from Sept 20, 2013, to Aug 24, 2018, from six local hospitals in Anhua County, Hunan Province, China. Mother-neonate pairs were eligible for inclusion if the neonates were born after Sept 20, 2013, and their mothers had resided in the study sites for at least 3 months. We tested samples with a haemagglutination inhibition (HAI) assay to measure antibody levels against three historical A(H1N1)pdm09 strains that were antigenically similar to the strains that circulated during the 2009 pandemic (A\/Hunan-Kaifu\/SWL4204\/2009 [SWL4204\/09 strain], A\/Hunan-Daxiang\/SWL1277\/2016 [SWL1277\/16 strain], and A\/Hunan-Yanfeng\/SWL185\/2018 [SWL185\/18 strain]). We also determined the seroprevalence, geometric mean titres (GMTs), transfer ratio of maternal antibodies, and the dynamics of maternally and naturally acquired antibodies in children, from birth to 3 years of age.<\/p>\n\n\n\n<p>\u65b9\u6cd5 \u57fa\u4e8e 2018 \u5e74 9 \u6708 20 \u65e5\u81f3 8 \u6708 24 \u65e5\u5728\u4e2d\u56fd\u6e56\u5357\u7701\u5b89\u5316\u53bf\u5efa\u7acb\u7684\u975e\u5e72\u9884\u6027\u7eb5\u5411\u961f\u5217\uff0c<br \/>\u672c\u7814\u7a76\u9009\u62e9\u4e86 528 \u540d\u961f\u5217\u513f\u7ae5\u53ca\u5176\u6bcd\u4eb2\u4f5c\u4e3a\u7814\u7a76\u5bf9\u8c61\u3002 \u91c7\u7528\u8840\u51dd\u6291\u5236\u5b9e\u9a8c\uff08haemagglutination<br \/>inhibition\uff0c HAI\uff09 \uff0c\u672c\u7814\u7a76\u68c0\u6d4b\u4e86\u4e0a\u8ff0\u7814\u7a76\u5bf9\u8c61\u5269\u4f59\u8840\u6e05\u6807\u672c\u4e2d\u7684\u7532\u578b H1N1 \u6d41\u611f\u75c5\u6bd2 A\/Hunan-<br \/>Kaifu\/SWL4204\/2009\uff08SWL4204\/09\uff09\u3001A\/Hunan-Daxiang\/SWL1277\/2016\uff08SWL1277\/16\uff09\u4ee5\u53ca<br \/>A\/Hunan-Yanfeng\/SWL185\/2018\uff08SWL185\/18\uff09\u6bd2\u682a\u7684 HAI \u6297\u4f53\u6c34\u5e73\u3002\u968f\u540e\uff0c\u7814\u7a76\u786e\u5b9a\u4e86\u5404\u6bd2<br \/>\u682a\u7684\u8840\u6e05\u9633\u6027\u7387\u3001\u6297\u4f53\u51e0\u4f55\u5e73\u5747\u6ef4\u5ea6\uff08geometric mean titre\uff0cGMT\uff09\u3001\u6bcd\u4f20\u6297\u4f53\u4f20\u8f93\u6bd4\u4ee5\u53ca\u513f<br \/>\u7ae5\u81ea\u51fa\u751f\u81f3 3 \u5468\u5c81\u671f\u95f4\u7684\u6bcd\u4f20\u548c\u81ea\u7136\u611f\u67d3\u8bf1\u5bfc\u6297\u4f53\u7684\u52a8\u6001\u53d8\u5316\u3002<\/p>\n\n\n\n<h3 class=\"wp-block-heading\">Findings<\/h3>\n\n\n\n<p>1066 mother-neonate pairs were enrolled in the original cohort between Sept 20, 2013, and Oct 14, 2015. Of these, 528 pairs (523 mothers, 528 neonates) were selected for the present study. The median age of the mothers was 25 years (IQR 23 to 29). 291 (55%) of 528 children were boys and 237 (45%) were girls, and most children (452 [86%]) were breastfed before the age of 6 months. The GMTs and the seroprevalence for the SWL4204\/09 strain were higher than those for the SWL1277\/16 and SWL185\/18 strains among mothers (GMTs: 10\u00b74 [95% CI 9\u00b78 to 11\u00b71]\u00a0<em>vs<\/em>\u00a09\u00b73 [8\u00b77 to 9\u00b78]\u00a0<em>vs<\/em>\u00a08\u00b70 [7\u00b75 to 8\u00b74], p&lt;0\u00b70001; seroprevalence: 11\u00b71% [95% CI 8\u00b75 to 14\u00b71]\u00a0<em>vs<\/em>6\u00b79% [4\u00b79 to 9\u00b74]\u00a0<em>vs<\/em>\u00a04\u00b76% [3\u00b70 to 6\u00b78], p=0\u00b70003) and among neonates (GMTs: 10\u00b77 [10\u00b70 to 11\u00b75]\u00a0<em>vs<\/em>\u00a09\u00b74 [8\u00b78 to 10\u00b70]\u00a0<em>vs<\/em>\u00a08\u00b71 [7\u00b76 to 8\u00b76], p&lt;0\u00b70001; seroprevalence: 13\u00b74% [10\u00b77 to 16\u00b77]\u00a0<em>vs<\/em>\u00a08\u00b77% [6\u00b75 to 11\u00b75]\u00a0<em>vs<\/em>\u00a06\u00b71% [4\u00b72 to 8\u00b75], p=0\u00b70002). Regardless of the A(H1N1)pdm09-specific strain, maternal antibodies could be transferred efficiently via the placenta (mean transfer ratios: 1\u00b710 for SWL4204\/09\u00a0<em>vs<\/em>1\u00b709 for SWL1277\/16\u00a0<em>vs<\/em>\u00a01\u00b706 for SWL185\/18; p=0\u00b793). The A(H1N1)pdm09 strain-specific antibodies waned below the protective threshold of 1:40 within 2 months after birth. After maternal antibody waning, there were periodic increases and decreases in HAI antibody titres against three A(H1N1)pdm09 strains, and such increases were all significantly associated with a higher immune response to heterologous strains. Vaccination against the SWL4204\/09 strain was associated with a poor response to the SWL185\/18 strain (\u03b2\u20130\u00b720, 95% CI \u20130\u00b728 to \u20130\u00b713; p&lt;0\u00b70001).<\/p>\n\n\n\n<p>\u7ed3\u679c 2013 \u5e74 9 \u6708 20 \u65e5\u81f3 2015 \u5e74 10 \u6708 14 \u65e5\uff0c \u961f\u5217\u5171\u7eb3\u5165 1066 \u540d\u513f\u7ae5\u53ca\u5176\u6bcd\u4eb2\uff0c\u5176\u4e2d 528<br \/>\u5bf9\u6bcd\u5a74\u5bf9 \uff08\u5305\u62ec 523 \u540d\u6bcd\u4eb2\u548c 528 \u540d\u65b0\u751f\u513f\uff09 \u88ab\u7eb3\u5165\u672c\u7814\u7a76\u3002 \u5728\u672c\u7814\u7a76\u4e2d\uff0c \u6bcd\u4eb2\u7684\u4e2d\u4f4d\u5e74\u9f84<br \/>\u4e3a 25 \u5c81\uff08IQR 23-29\uff09\uff1b\u7537\u5b69\u4e3a 291 \u540d\uff0855%\uff09\uff0c\u5973\u5b69\u4e3a 237 \u540d\uff0845%\uff09\uff1b452 \u540d\uff0886%\uff09\u513f<br \/>\u7ae5\u5728 6 \u6708\u9f84\u524d\u63a5\u53d7\u4e86\u6bcd\u4e73\u5582\u517b\u3002\u7814\u7a76\u53d1\u73b0\uff0c\u6bcd\u4eb2\uff08GMT\uff1a10.4 [95% CI\uff1a9.8-11.1]\uff0c9.3 [8.7-<br \/>9.8]\uff0c8.0 [7.5-8.4]\uff0cp&lt;0.0001\uff1b\u8840\u6e05\u9633\u6027\u7387\uff1a11.1% [95% CI\uff1a8.5-14.1]\uff0c6.9% [4.9-9.4]\uff0c4.6%<br \/>[3.0-6.8]\uff0c p=0.0003\uff09\u548c\u65b0\u751f\u513f\uff08GMT\uff1a 10.7 [10.0-11.5]\uff0c 9.4 [8.8-10.0]\uff0c 8.1 [7.6-8.6]\uff0c p&lt;0.0001\uff1b<br \/>\u8840\u6e05\u9633\u6027\u7387\uff1a13.4% [10.7-16.7]\uff0c8.7% [6.5-11.5]\uff0c6.1% [4.2-8.5]\uff0cp=0.0002\uff09\u7684 SWL4204\/09<br \/>\u682a\u7684 GMT \u548c\u8840\u6e05\u9633\u6027\u7387\u5747\u9ad8\u4e8e SWL1277\/16 \u548c SWL\/185\/18 \u682a\u3002\u4e0d\u8bba\u4f55\u79cd\u6bd2\u682a\uff0c\u6bcd\u4eb2\u6297\u4f53\u5747<br \/>\u53ef\u901a\u8fc7\u80ce\u76d8\u6709\u6548\u4f20\u8f93\u7ed9\u65b0\u751f\u513f \uff08\u5e73\u5747\u4f20\u8f93\u6bd4\uff1a SWL4204\/09 \u682a\u4e3a 1.10\u3001 SWL1277\/16 \u682a\u4e3a 1.09\u3001<br \/>SWL185\/18 \u682a\u4e3a 1.06\uff0cp=0.93\uff09\u3002\u7814\u7a76\u8fd8\u53d1\u73b0\uff0c\u5404\u6bd2\u682a\u7684 HAI \u6297\u4f53\u5728\u513f\u7ae5\u51fa\u751f\u540e 2 \u4e2a\u6708\u5185\u8870<br \/>\u51cf\u5230\u4fdd\u62a4\u9608\u503c 1:40 \u4ee5\u4e0b\uff1b\u6bcd\u4f20\u6297\u4f53\u8870\u51cf\u540e\uff0cHAI \u6297\u4f53\u6ef4\u5ea6\u5448\u73b0\u5468\u671f\u6027\u5347\u9ad8\u548c\u4e0b\u964d\uff0c\u5e76\u4e14\u5404<br \/>\u6bd2\u682a\u7684\u6297\u4f53\u6ef4\u5ea6\u7684\u5468\u671f\u6027\u5347\u9ad8\u4e0e\u5f02\u79cd\u6bd2\u682a\u6297\u4f53\u6ef4\u5ea6\u7684\u5347\u9ad8\u663e\u8457\u76f8\u5173\uff1b\u9488\u5bf9 SWL4204\/09 \u682a\u7684<br \/>\u75ab\u82d7\u63a5\u79cd\u4e0e SWL185\/18 \u682a\u6297\u4f53\u6ef4\u5ea6\u7684\u964d\u4f4e\u6709\u5173\uff08\u03b2=-0.20\uff0c 95%CI\uff1a-0.28 \u81f3-0.13\uff0c p\uff1c0.0001\uff09\u3002<\/p>\n\n\n\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" src=\"https:\/\/ars.els-cdn.com\/content\/image\/1-s2.0-S2666524723001817-gr1_lrg.jpg\" alt=\"\"\/><\/figure>\n\n\n\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" src=\"https:\/\/ars.els-cdn.com\/content\/image\/1-s2.0-S2666524723001817-gr2_lrg.jpg\" alt=\"\"\/><\/figure>\n\n\n\n<figure class=\"wp-block-image size-large\"><img decoding=\"async\" src=\"https:\/\/ars.els-cdn.com\/content\/image\/1-s2.0-S2666524723001817-gr3_lrg.jpg\" alt=\"\"\/><\/figure>\n\n\n\n<h3 class=\"wp-block-heading\">Interpretation<\/h3>\n\n\n\n<p>Our findings suggest low pre-existing immunity against influenza A(H1N1)pdm09 virus among unvaccinated Chinese adult female and paediatric populations.
