Effect of Hydroxychloroquine in Hospitalized Patients with COVID-19: Preliminary results from a multi-centre, randomized, controlled trial<\/h1>\n\n\n\n

Hydroxychloroquine and chloroquine have been proposed as treatments for coronavirus disease 2019 (COVID-19) on the basis of in vitro activity, uncontrolled data, and small randomized studies.<\/p>\n\n\n\n

\u6839\u636e\u4f53\u5916\u6d3b\u6027\uff0c\u975e\u5bf9\u7167\u7814\u7a76\u6570\u636e\u4ee5\u53ca\u5c0f\u6837\u672c\u968f\u673a\u8bd5\u9a8c\uff0c\u7f9f\u6c2f\u55b9\u548c\u6c2f\u55b9\u88ab\u7528\u4e8e\u6cbb\u7597\u65b0\u51a0\u75c5\u6bd2\u611f\u67d3(COVID-19)\u3002<\/p>\n\n\n\n

Methods \u65b9\u6cd5<\/h3>\n\n\n\n

The Randomised Evaluation of COVID-19 therapy (RECOVERY) trial is a randomized, controlled, open-label, platform trial comparing a range of possible treatments with usual care in patients hospitalized with COVID-19. We report the preliminary results for the comparison of hydroxychloroquine vs. usual care alone. The primary outcome was 28-day mortality.<\/p>\n\n\n\n

RECOVERY\u8bd5\u9a8c\u662f\u4e00\u9879\u968f\u673a\u3001\u5bf9\u7167\u3001\u5f00\u653e\u6807\u7b7e\u3001\u5e73\u53f0\u8bd5\u9a8c\uff0c\u6bd4\u8f83\u6570\u79cd\u6cbb\u7597\u4e0e\u5e38\u89c4\u6cbb\u7597\u5bf9\u4e8eCOVID-19\u4f4f\u9662\u60a3\u8005\u7684\u7597\u6548\u3002\u6211\u4eec\u62a5\u544a\u7f9f\u6c2f\u55b9\u4e0e\u5e38\u89c4\u6cbb\u7597\u8fdb\u884c\u6bd4\u8f83\u7684\u521d\u6b65\u7ed3\u679c\u3002\u4e3b\u8981\u9884\u540e\u7ec8\u70b9\u4e3a28\u5929\u75c5\u6b7b\u7387\u3002<\/p>\n\n\n\n

Results \u7ed3\u679c<\/h3>\n\n\n\n

1561 patients randomly allocated to receive hydroxychloroquine were compared with 3155 patients concurrently allocated to usual care. Overall, 418 (26.8%) patients allocated hydroxychloroquine and 788 (25.0%) patients allocated usual care died within 28 days (rate ratio 1.09; 95% confidence interval [CI] 0.96 to 1.23; P=0.18). Consistent results were seen in all pre-specified subgroups of patients. Patients allocated to hydroxychloroquine were less likely to be discharged from hospital alive within 28 days (60.3% vs. 62.8%; rate ratio 0.92; 95% CI 0.85-0.99) and those not on invasive mechanical ventilation at baseline were more likely to reach the composite endpoint of invasive mechanical ventilation or death (29.8% vs. 26.5%; risk ratio 1.12; 95% CI 1.01-1.25). There was no excess of new major cardiac arrhythmia.<\/p>\n\n\n\n

Conclusions \u7ed3\u8bba<\/h3>\n\n\n\n

In patients hospitalized with COVID-19, hydroxychloroquine was not associated with reductions in 28-day mortality but was associated with an increased length of hospital stay and increased risk of progressing to invasive mechanical ventilation or death.<\/p>\n\n\n\n
